The C-terminal amino acid was supplied coupled to a PEG-PS support and the amino acids were sequentially coupled after removal of the Fmoc-blocking group at each cycle. The reagents used were 20% piperidine in DMF for the de-blocking and HATU for the coupling steps as recommended by the manufacturer. The peptide was cleaved and deprotected from the PEG-PS support by treating with TFA reagent (1.76 mL TFA, 100 mg DTT, 40 L triisopropylsilane and 100 L water) for approximately 4 hours followed by precipitation and partial purification with tert-butylmethylether.The crude peptide (74 mg) was dissolved in 6 mL of 0.1% TFA/10% acetonitrile (v/v) in water and purified by HPLC on a Jupiter Proteo column (10 x 250 mm, RP-C12, 4 m particle size, 90-angstrom pore size, Phenomenex) connected to an Agilent 1100 series binary pump and monitored using an Agilent photodiode array detector. Detection was carried out at 214 nm. Purification was first carried out using a gradient (0 % to 100 % v/v over 1 hr) of solvent B [0.085 % TFA /70 % acetonitrile (v/v) in water] in solvent A [0.1 % TFA (v/v) in water] with a flow rate of 2 mL/min. Three major fractions (A, B and C) of repeated injections (500 μL) were collected and concentrated under vacuum at 4 degrees C.
